Is there really a big difference between 2000's and 2001+'s , i know the interior looks different between 2000 and ups but are there any really significant changes that would merit wanting a 2001+ over a 2000?

I believe that the 2000 Durango R/T required premium fuel but they changed it during model year 2001 so later models can run regular. My 01 does not require premium, but I believe that my owners manual mentions that some did.
